ABSTRACT

S-adenosyl-l-methionine (SAM) is ubiquitous in living organisms and plays important roles in transmethylation, transsulfuration and transamination in organisms. Due to its important physiological functions, production of SAM has attracted increasing attentions. Currently, researches on SAM production mainly focus on microbial fermentation, which is more cost-effective than that of the chemical synthesis and the enzyme catalysis, thus easier to achieve commercial production. With the rapid growth in SAM demand, interests in improving SAM production by developing SAM hyper-producing microorganisms aroused. The main strategies for improving SAM productivity of microorganisms include conventional breeding and metabolic engineering. This review summarizes the recent research progress in improving microbial SAM productivity to facilitate further improving SAM productivity. The bottlenecks in SAM biosynthesis and the solutions were also addressed.

ABSTRACT

Extracellular vesicles (EVs), also known as membrane vesicles, are vesicular bodies secreted by eukaryotic cells and bacteria. EVs can carry proteins, DNA, RNA, and various metabolites for the exchange and transmission of substances between cells. They play contents-dependent physiological functions, such as delivering nutrients, participating in immune response, and treating cancers. Currently, most studies focus on the exploration of vesicles secreted by eukaryotic cells and gram-negative bacteria, while few studies focus on gram-positive bacteria. This review summarized the production, content composition, physiological function, and engineering of EVs secreted by gram-positive bacteria, and prospected future perspectives in this area.

ABSTRACT

Objective:To investigate the promoting effect of Difrarel ? on retinal function following panretinal photocoagulation (PRP) in the eyes with diabetic retinopathy. Methods:A non-randomized controlled study was performed.A total of 108 eyes of 108 patients with non-proliferative diabetic retinopathy (NPDR) were enrolled in Tongren Ophthalmology Center and Beijing Daxing District People's Hospital from December 2014 to February 2020.The patients were divided into PRP group and PRP+ Difrarel ? group according to different therapies under patients' selection.Difrarel ? was orally administered after PRP in 56 patients of PRP+ Difrarel ? group, and only PRP was given in 52 patients of PRP group.The visual acuity, 30°~60° circular visual field and multifocal electroretinogram were examined before and 1 day, 1 month, 3 months, 6 months, 12 months after PRP.The central macular thickness (CMT) was measured by optical coherence tomography, and fundus neovascularization was observed by fluorescein fundus angiography at 6 and 12 months after PRP.The study protocol was approved by an Ethics Committee of Beijing Daxing District People's Hospital (No.2021-F4). Results:Visual improvement rate was 57.14% (32/56) and 32.69% (17/52) in PRP+ Difrarel ? group and PRP group at the end of following-up, respectively, showing a significant difference between two groups ( χ2=3.56, P<0.05). The visual field mean sensitivity was significantly different at different time points in two groups ( Fgroup=4.77, P<0.05; Ftime=6.51, P<0.05), and was lower after PRP than those before treatment in both groups (both at P<0.05), and was significantly higher in PRP+ Difrarel ? group than PRP group at 3, 6, 12 months after PRP (all at P<0.05). The P1 amplitude density in 3 to 5 rings in PRP+ Difrarel ? group were higher than those in PRP group, and the differences were statistically significant (all at P<0.05). There was no significant difference in CMT between the two groups at different time points ( Fgroup=3.57, P>0.05; Ftime=1.23, P>0.05). No new blood vessels and non-perfusion area were found in both groups. Conclusions:Oral Difrarel ? can improve retinal function after PRP in the eyes with NPDR.

ABSTRACT

Transient receptor potential vanilloid 3 (TRPV3) is a non-selective cation channel, located on cell membranes. TRPV3 is extensively expressed in various organs such as skin, brain, dorsal root ganglia, heart and colon. It's reported that TRPV3 involves in many physiological processes including sensation, skin barrier formation, hair growth and vasodilatation, and pathological processes like pruritus, cutaneous inflammatory disease and cancer. TRPV3 can respond to innoxious warm stimulation (≥ 33 ‍℃‍), endogenous substances (e.‍g., farnesylpyrophosphate) and exogenous small molecules (e.g., carvacrol, camphor and 2-APB). Recently, several natural or synthetic small molecules (e.g., osthole, 74a and dyclonine) have been shown to suppress TRPV3 activity, accompanying with therapeutic efficacy in animal models of diseases, which suggests the potential of TRPV3 as drug target. This paper reviews the research progress on the structure, physiological functions, related diseases and modulators of the TRPV3 channel to provide theoretical references for the future study on TRPV3 channel.

■ RESUMO Os defeitos na região palpebral são causados principalmente por excisões cirúrgicas de neoplasias cutâneas. Os objetivos da reconstrução palpebral estão fundamentados na restauração da funcionalidade desta unidade anatômica para manter a proteção ocular e a recuperação de uma aparência normal devido à importância crítica da região periocular na estética facial. O reparo dos defeitos palpebrais começa com uma cuidadosa avaliação dos componentes anatômicos que têm sido ressecados e precisam ser reconstruídos; a extensão e a localização do defeito guiarão a reconstrução. Grandes defeitos comprometendo a totalidade da espessura palpebral são um desafio para os cirurgiões plásticos. Milhares de técnicas cirúrgicas têm sido descritas para a reconstrução de defeitos palpebrais de espessura total; apresentamos neste artigo a descrição de um caso de reconstrução dinâmica da pálpebra com associação de um retalho frontal com transposição do músculo temporal após ressecção de um carcinoma basocelular infiltrativo recidivado.


■ ABSTRACT Surgical excisions of skin neoplasms mainly cause defects in the eyelid region. The objectives of eyelid reconstruction are based on the restoration of this anatomical unit's functionality to maintain eye protection and recovery from a normal appearance due to the critical importance of the periocular region in facial aesthetics. Repair of eyelid defects begins with a careful evaluation of the anatomical components that have been resected and need to be reconstructed; the extent and location of the defect will guide the reconstruction. Large defects compromising the entire body thickness are a challenge for plastic surgeons. Thousands of surgical techniques have been described for the reconstruction of total thickness eyelid defects; we present in this article the description of a case of dynamic eyelid reconstruction with an association of a frontal flap with temporal muscle transposition after resection of a recurrent infiltrative basal cell carcinoma.

ABSTRACT

Proteases are widely found in organisms participating in the decomposition of proteins to maintain the organisms' normal life activities. Protease inhibitors regulate the activities of target proteases by binding to their active sites, thereby affecting protein metabolism. The key amino acid mutations in proteases and protease inhibitors can affect their physiological functions, stability, catalytic activity, and inhibition specificity. More active, stable, specific, environmentally friendly and cheap proteases and protease inhibitors might be obtained by excavating various natural mutants of proteases and protease inhibitors, analyzing their key active sites by using protein engineering methods. Here, we review the studies on proteases' key active sites and protease inhibitors to deepen the understanding of the active mechanism of proteases and their inhibitors.

ABSTRACT

Idiopathic pulmonary fibrosis (IPF) is a chronic fatal pulmonary disease characterized by complex illness condition. There is no effective treatment at present except lung transplantation. The comprehensive evaluation is helpful for the management of patients with IPF in hierarchical stages. Therefore, it is very important to evaluate IPF by various independent factors. At present, the commonly used methods for clinical evaluation on IPF include assessment of health-related quality of life, assessment of physiological function, assessment of imaging, assessment of laboratory examination, and multi-dimensional assessment system. However, there are different advantages and disadvantages on diverse evaluation methods for the evaluation of IPF.

@#As a key component of glutamatergic system, metabotropic glutamate receptor 5 (mGluR5) has been extensively involved in the regulation of physiological processes such as synaptic transmission, synaptic plasticity and synaptic excitation/inhibition balance.Over the past few decades, mGluR5 has been found to be closely related to multiple neurological and psychiatric disorders, thus it is of considerable interest as a drug target in the treatment of such disorders.This review summarizes the structure and distribution of mGluR5, its normal physiological function, its pathological roles in related central nervous system (CNS) diseases, as well as the current status of its drug development, in order to provide reference for further investigation.

Abstract Introduction: Cancer of the oral cavity and oropharynx presents aggressive behavior and its diagnosis is, in most cases, performed in advanced stages. Total glossectomy is a therapeutic option in locally advanced cancer, and the only one in the recurrent or residual disease, after chemoradiotherapy. Objective: To evaluate the clinical-epidemiological profile, postoperative complications, survival rates and functional aspects of patients with oral cavity and oropharynx cancer after total glossectomy. Methods: It was a retrospective study where 22 patients were included with oral cavity and oropharyngeal cancer after total glossectomy at the Hospital Regional do Vale do Paraíba, em Taubaté, São Paulo. Results: All patients were male, with a median age of 57 years, most of tumors are located in the tongue and floor of the mouth and classified as stage IVa. Total glossectomy as initial treatment was performed in 18 and as salvage in four patients. The major pectoralis myocutaneous flap was used for reconstruction in all cases. The main postoperative complication was wound infection and salivary fistula. Conclusion: Overall survival was 19% and cancer-specific survival was 30.8% in five years. Eight patients were rehabilitated for exclusive oral feeding without the dependence tracheostomy and enteral tube, all with an overall survival greater than 15 months.


Resumo Introdução: O câncer da cavidade oral e da orofaringe apresenta comportamento agressivo e seu diagnóstico é, na maioria dos casos, realizado em fases avançadas. A glossectomia total é uma opção terapêutica no câncer localmente avançado e a única no resgate de pacientes com doença residual ou recorrente, após a quimiorradioterapia. Objetivo: Avaliar o perfil clínico-epidemiológico, as complicações pós-operatórias, as taxas de sobrevida e os aspectos funcionais de pacientes com câncer da cavidade oral e da orofaringe submetidos à glossectomia total. Método: Estudo retrospectivo em que foram incluídos 22 pacientes com câncer de cavidade oral e orofaringe submetidos à glossectomia total no Hospital Regional do Vale do Paraíba, em Taubaté, São Paulo. Resultados: Todos os pacientes eram do gênero masculino, com mediana de 57 anos, com tumores principalmente na língua e no assoalho da boca e classificados, em sua maioria, como estádio clínico IVa. A glossectomia total como tratamento inicial foi realizada em 18 e como resgate em quatro pacientes. O retalho miocutâneo peitoral maior foi utilizado para a reconstrução em todos os casos. A principal complicação pós-operatória foi a infecção da ferida operatória e a fístula salivar. Conclusão: A sobrevida global foi de 19% e a específica por câncer de 30,8% em cinco anos. Oito pacientes foram reabilitados para alimentação oral exclusiva sem a dependência de traqueostomia e ou de dieta enteral, todos com sobrevida global maior do que 15 meses.

Abstract Rhizosphere microorganisms and endophytes can help their hosts absorb nutrients and regulate the levels of plant hormones. Moreover, they can modulate the expressions of host genes, assist hosts in eliminating reactive oxygen species (ROS) and secreting volatile organic compounds. Therefore, rhizosphere microorganisms and endophytes are considered as determinant factors driving processes involved in the growth of host plants. However, the physiological and ecological functions, as well as the molecular mechanism underlying the behavior of rhizosphere microorganisms and endophytes in their role in the adaptive capacity of host plants in the karstic high-calcium environment have not been systematically studied. This review summarizes the physiological and molecular mechanisms of rhizosphere microorganisms and endophytes which help host plants to adapt to various kinds of adverse environments. The adaptive capacities of plants growing in adverse environments, partly, or totally, depends on microorganisms co-existing with the host plants.


Resumo Os microorganismos e endófitos da rizosfera podem ajudar seus hospedeiros a absorver nutrientes e regular os níveis de hormônios vegetais. Além disso, eles podem modular as expressões dos genes hospedeiros, auxiliar os hospedeiros na eliminação de espécies reativas de oxigênio (ROS) e secretar compostos orgânicos voláteis. Portanto, microorganismos e endófitos da rizosfera são considerados determinantes dos processos envolvidos no crescimento de plantas hospedeiras. No entanto, as funções fisiológicas e ecológicas, bem como o mecanismo molecular subjacente ao comportamento dos microrganismos e endofíticos da rizosfera no seu papel na capacidade adaptativa das plantas hospedeiras no ambiente cárstico de alto teor de cálcio, não foram sistematicamente estudados. Esta revisão resume os mecanismos fisiológicos e moleculares de microrganismos e endófitos da rizosfera que ajudam as plantas hospedeiras a se adaptarem a vários tipos de ambientes adversos. As capacidades adaptativas das plantas que crescem em ambientes adversos, em parte ou totalmente, dependem de microrganismos coexistentes com as plantas hospedeiras.

ABSTRACT

Nanogenerator (triboelectric nanogenerator and piezoelectric nanogenerator) has experienced a rapid development since it was proposed. This technique can covert various mechanical energies into electric energy, including human motion energy, wind energy, acoustic energy and ocean energy. The converted electricity can be used for health monitoring and physiological function regulation, such as pulse detection, bioelectrical stimulation and cardiac pacing. This review summarizes the structure, working mechanism, output performance of nanogenerator and its latest progress in circulatory system, nervous system, biological tissue, sleep and rescue system. Additionally, a further analysis was also made on the application challenge of nanogenerator in clinical treatment. In the future, nanogenerator is expected to be an auxiliary power source, or even to replace battery to power medical electronic device and realize the self-powered health monitoring and physiological function regulation of human body.

ABSTRACT

The α1-adrenergic receptor (α1-AR) belongs to G-protein coupled receptors (GPCRs) and contains three subtypes, namely α1A, α1B and α1D. Studies have shown that structural changes in the α1-ARs receptor can lead to changes in their physiological functions. In the present study, we summarize the structural changes in four aspects of α1-ARs receptors, including carboxy-terminal truncation, dimerization, allosteric effects, and point mutations that lead to endocytosis, phosphorylation, desensitization, and affinity for agonists.

ABSTRACT Objective: To assess the clinical and functional results of patients submitted to reverse arthroplasty with a minimum follow-up of one year. Methods: Twenty-two patients submitted to shoulder reverse arthroplasty by the Surgery and Shoulder Rehabilitation Group were retrospectively evaluated with pre and postoperative imaging analysis, analog pain scale, range of motion, and ASES functional score. Results: Out of 19 (86.3%) patients with preoperative ASES classified as poor/bad, 11 (57.9%) progress to good/excellent after intervention, showing improvement of function, ranging from a mean preoperative ASES score of 22 (± 18.8) to a postoperative mean of 64.8 (± 27.7) (p = 0.031). Regarding the pain, there was an improvement in analog pain scale, presenting a preoperative mean of 7.64 (1-10) and a postoperative mean of 2.09 (0-7; p < 0.001). Regarding mobility, of 22 patients, 15 (68.2%) had preoperative pseudoparalysis and, of these, ten (66.7%) had an active anterior elevation greater than 90° after reverse arthroplasty. In turn, patients without pseudoparalysis had no significant gain in range of motion (p = 0.002). The authors observed active anterior elevation gain, with a preoperative mean of 76° (0-160°) and a postoperative mean of 111° (0-160°; p = 0.002). Conclusion: Despite being a relatively new procedure in Brazil, reverse shoulder arthroplasty can be used effectively and safely in patients who were previously without treatment options such as rotator cuff arthropathy and revisions providing pain relief, improvement of function, and mobility of the upper limb.


RESUMO Objetivo: Avaliar os resultados clínicos e funcionais de pacientes submetidos a artroplastia reversa com seguimento mínimo de um ano. Métodos: Foram avaliados retrospectivamente 22 pacientes submetidos a artroplastia reversa de ombro pelo grupo de cirurgia e reabilitação de ombro da nossa instituição com análise pré e pós-operatória de exames de imagem, escala analógica da dor, amplitude de movimento e escala funcional ASES. Resultados: Dos 19 (86,3%) pacientes que apresentavam ASES pré-operatória classificada como péssimo/ruim, 11 (57,9%) evoluíram para bom/excelente após a intervenção, apresentaram melhoria da função, saíram de uma escala ASES pré-operatória média de 22 (±18,8) para uma pós-operatória de 64,8 (± 27,7; p = 0,031). Quanto à dor, observou-se melhoria da escala analógica da dor, apresentaram média pré-operatória de 7,64 (1-10) e pós-operatória de 2,09 (0-7; p < 0,001). Em relação à mobilidade, dos 22 pacientes, 15 (68,2%) apresentavam pseudoparalisia pré-operatória; desses, dez (66,7%) passaram a apresentar elevação anterior ativa superior a 90° após artroplastia reversa. Por outro lado, os pacientes sem pseudoparalisia não apresentaram ganho significativo de amplitude de movimento (p = 0,002). Foi observado ganho de elevação anterior ativa, com média pré-operatória de 76° (0-160°) e pós-operatória de 111° (0-160°; p = 0,002). Conclusão: Apesar de ser um procedimento relativamente novo no Brasil, a artroplastia reversa de ombro pode ser usada com eficácia e segurança em pacientes que previamente apresentavam-se sem opções terapêuticas como artropatia do manguito rotador e revisões que proporcionam alívio de dor, melhoria da função e mobilidade do membro superior.

ABSTRACT

Objective To evaluate the effect of type WIYIS-6/90 plateau oxygen generating vehicle on the high altitude spot. Methods Totally 20 male soldiers during field training at 4 280 m altitude were involved in this trial. They were given administration of oxygen inhalation at 2 L/min by nasal cannula. Blood pressure, heart rate, blood oxygen saturation, vital capacity, left and right hand strength, digit span and discriminative reaction time were detected before and after oxygen inhalation for 20 minutes and finished for 10 minutes. Results After oxygen inhalation for 20 minutes, blood oxygen saturation, vital capacity, left and right hand strength, digit span and correct times increased and heart rate decreased compared with before oxygen inhalation (P<0.05,or P<0.01).Stopping oxygen inhalation for 10 minutes,vital capacity and right hand strength increased and response time decreased(P<0.05,or P<0.01).Conclusion Using type WIYIS-6/90 plateau oxygen generating vehicle for oxygen supply at high altitude can improve brain-body function and promote work ability.

Objective To investigate the short-term efficacy of treating thyroid multiple-nodule by Ultrasound-guided microwave ablation in one-session,and to evaluate its safety and effect.Methods Treatment and follow-up data of 85 patients with benign thyroid nodules underwent tUltrasound-guided microwave ablation in one-session were retrospectively analyzed.Results Symptom scores and cosmetic scores increased with the increasing number of nodules ablated before ablation (P < 0.001),as well as the ablation time (P < 0.001).However,there was no significant difference in the amount of isolation belt and intraoperative pain score between the groups.There were no serious complications such as recurrent laryngeal nerve injury and massive hemorrhage in 85 cases underwent tUltrasound-guided microwave ablation in one-session.Compared with the thyroid function,there was no significant difference on before the ablation and 1,3,6 months after ablation.Postoperative 6 months,the maximum volume of nodule and the total volume of the nodules were reduced 63% and 69.5%,respectively.After ablation,both the symptom scores and cosmetic scores were improved significantly.On 6 months after ablation,the symptom scores decreased from (4.41 ± 1.75) cm to (1 ± 0.76) cm,and the cosmetic score3 decreased from 2.27 ± 0.81 to 1.25 ± 0.43.Conclusion Ultrasound-guided microwave ablation could treat multiple thyroid nodules effectively and protect the physiological function of thyroid,which was worthy of further promotion in clinical application.

They play an important role in the regulation of many physiological and pathological processes through regulating embryonic develop-ment,cell growth,differentiation,cell apoptosis and interac-tions of cells under the regulation of MAP kinases(ERK,p38 and JNK),Ca2 + relevant signaling pathways and TGF-β path-ways. Notably,many Ets members involve in tumor genesis,in-vasion and metastasis,and these members may be markers of anticipating tumor genesis,development and prognosis. Here the physiological role of Ets family members and its relationship with human diseases are summarized,aiming to provide a theoretical basis for the study of the role of Ets family in human diseases.

ABSTRACT

The NOD protein family include NOD1 and NOD2,both of which recognize intracellular bacterial peptidoglycan,induce inflammation and promote host defense.In this review article,we summarize the research progress on NOD family,including signaling and regulation.We will also discuss their cellular localization and the role in host defense.Understanding on their physiological function will help shed light on pathogenesis on related diseases.
